ThePrincipal Researcher, IHC will contribute to our discovery biology efforts by conducting a wide variety of immunohistochemical studies on tissues from animal models and humans. This person will be able to work independently and have sufficient background to be able to clearly present his/her experimental rationale, data, and analysis. Responsibilities include preparing tissues for sectioning, generating tissue sections by microtomy and cryotomy, designing, validating, and performing manual and automated immunohistochemistry, immunofluorescence and in situ hybridization experiments, imaging resulting slides by microscope or slide scanner, and analyzing the resulting images. Experience in neuroscience/ neuroinflammation is preferred. The ideal candidate should be well organized, proactive, possess excellent technical and communication skills, as well as the ability to work collaboratively in a team environment. Flexibility and willingness to learn and master new techniques are critical. We seek a creative and diligent person who brings care and attention to detail to the job.
Essential Functions
* Independently develop, optimize, and validate protocols for single and multiplexed immunohistochemistry, immunofluorescence and in-situ hybridization
* Prepare fixed tissue for sectioning
* Section frozen and paraffin embedded tissue samples and perform routine histology staining procedures
* Perform both manual and automated staining protocols
* Acquire images of stained sections using microscope (confocal, epifluorescence) or slide scanner
* Independently perform image analysis and quantification
* Generate accurate and detailed study reports and maintain other necessary records including a lab notebook.
* Independently identify and troubleshoot experimental roadblocks
* Participate in project team meetings
* Maintain laboratory equipment and reagent inventory
* Work well in a team environment
Requirements
* M.S. or B.S. plus 4-6 years relevant experience in Neuroscience, Pharmaceutical Sciences or other related field
* A background in neuroimmunology and/ or neurodegeneration is preferred
* Extensive experience with tissue embedding, tissue sectioning (microtomy and cryotomy), and manual immunohistochemistry and immunofluorescence staining is required
* Extensive experience with confocal, epifluorescence, and bright field microscopy required
* Experience with digital slide scanners (e.g. Leica Versa) preferred
* Experience with automated staining platforms (e.g. Leica BondRx and/or Ventana Discovery Ultra) preferred
* Experience with image analysis software (Imaris, Halo, Matlab, etc) strongly preferred
* Experience with pre-clinical species (e.g. mouse) required and experience with mouse brain sectioning/ staining preferred
* Excellent communication skills
* Detail oriented and highly motivated
