The Vertex Summer Co-op Program offers students the opportunity to learn about the pharmaceutical industry from a hands-on perspective. Generation and functional characterization of iPSC-derived skeletal muscle cells (iSKMs) amenable for gene editing
Abstract
Vertex Pharmaceuticals is working to discover and develop innovative genetic therapies with the potential to provide transformative benefit to patients. Vertex is currently advancing multiple CRISPR-based genetic therapies that have the potential to cure or stop the progression of degenerative and neuromuscular diseases. This internship will provide a fantastic opportunity to work on cutting edge science, apply state-of-the-art gene editing technologies, and take part in discovering and developing next-generation therapies for Duchenne Muscular Dystrophy, Myotonic Dystrophy Type 1 and other genetic disorders.
The internship will be undertaken at Vertex Cell and Genetic Therapies, in a highly dynamic and competitive environment. The overarching goal of this research is to evaluate gene editing efficiency at multiple disease genomic loci and to create an in vitro model system for screening and delivery of next-generation therapeutic approaches. The proposed project will establish robust in vitro functional assays for the characterization of muscle cells following differentiation of patient-derived iPSCs. We will initially focus on the characterization of muscle fiber maturity through analysis of a panel of muscle markers, as well as methods for therapeutic asset delivery and functional readouts in muscle cells. The successful candidate will be exposed to a broad range of advanced technologies including CRISPR-Cas9 genome engineering, iPS cell culture and differentiation systems, and advanced microscopy imaging.
