Wave Life Sciences USA, Inc. seeks Scientists I, Biology for Cambridge, MA offices. Duties include: Design and conduct lab experiments for optimization of ASO’s in CNS relevant cell models, including in vitro activity evaluation and IC50 assessment, and structure activity relationship analysis; Develop siRNA platform and investigate impacts of chemistry and stereochemistry in in vitro cellular uptake and activity in CNS relevent cell models; Explore optimized chemistry optimization patterns of siRNA for productive delivery to various extra-hepatic primary cell models. Position requires a MS, or foreign equivalent, in Pharmaceutical Sciences, Bioengineering, Biomedical Sciences, Biology, or related field and 4 years’ experience in nucleic acid therapeutic research and development, or alternatively, a BS degree or foreign equivalent in Pharmaceutical Sciences, Bioengineering, Biomedical Sciences, Biology, or related field and 6 years’ experience in nucleic acid therapeutic research and development.
Experience must include the following, which may be gained concurrently:
• 4 years’ experience and expertise in discovery and research of oligonucleotide therapeutics, including 4 years’ experience using in-vitro models, and 2 years using in-vivo models relevant to CNS diseases;
• 4 years’ experience in platform development and mechanistic studies to improve the efficacy and potency of oligonucleotide therapeutics, including 4 years’ experience with ASOs and 2 years’ experience with siRNAs;
• 4 years’ experience drafting and revising Standard Operating Protocols (SOPs) for biological studies and other important documents with in-vitro sample processing for transcript and protein study, and 2 years’ experience drafting and revising SOPS with in-vivo sample processing for transcript and protein study;
• 4 years’ experience in qualitative and quantitative analysis of ASO mediated studies involving High-throughput screening (in-vitro);
• 3 years’ experience evaluating supplies and equipment to ensure operability in laboratory settings including High throughput Liquid handlers like RNA extraction kits and reagents (like enzymes, buffers), thermocyclers, and plate readers;
• 2 years’ experience evaluating supplies and equipment to ensure operability in laboratory settings including High throughput Liquid handlers like-Agilent Bravo, Formulatrix Mantis, Biotek Plate washers, and Tissue disruptor system like Tissue pulverizer and its components;
• 2 years’ experience in research and development of siRNA therapeutics for both common and rare hepatic diseases including chronic liver disease, metabolic disorders, inborn error, and metabolism;
• 2 years’ experience in qualitative and quantitative analysis with siRNA mediated studies involving High-throughput screening, PK/PD, toxicity, and target engagement (in-vivo);
• 2 years’ experience in culture and maintenance of primary cells for different extra-hepatic tissues;
• 2 years’ experience in generation and optimization of iPSC derived neurons and maintenance and culture of primary cells;
• 2 years’ experience in siRNA platform development for extra-hepatic tissue distribution and productive delivery; and
• 2 years’ experience in processing and analysis of cells and tissue samples from in-vivo studies applying an understanding in-vivo pharmacology of oligonucleotides.
