Background:The Giraldez Lab at Yale School of Medicine is seeking a postgraduate associate interested in uncovering the dynamic mechanisms underlying zygotic genome activation (ZGA) in the context of developing zebrafish embryos. The regulation of many genes activated during ZGA is accomplished through the actions of pioneer transcription factorswhich facilitate chromatin accessibility and histone acetylation. We will use live imaging and expansion microscopy to investigate the dynamics of ZGA. These approaches are complementary, enabling high temporal resolution measurements via live imaging and high spatial resolution measurements via expansion microscopy. Training:The new hire will receive training in Genetics,Molecular and Developmental Biology using the Zebrafish model. This will include being trained on several necessary lab techniques including CRISPR, DNA extraction, cloning, general zebrafish handling and crossing, zebrafish tissue collection, genotyping using PCR and gel electrophoresis, expansions microscopy (ChromExM), fluorescence microscopy, MS2-MCP imaging of live gene expression, and data interpretation and analysis. Additionally, they will be trained lab organization, documentation, and record keeping as well as how to document and present results using Illustrator, Photoshop, and PowerPoint.The postgraduate associate will be mentored byassigned members of the Giraldez lab to gain experience in experimental design, execution and analysis of experiments, lab meeting presentation and time management.
Qualifications/Education:The ideal candidate should be a recent graduate with a Bachelor’s degree in Biology or a biological science with an intent to apply to graduate school within the next two years. They should be highly motivated and dedicated to high level research and ideally already have experience in research involving molecular biology and/or fluorescence microscopy.
